Mode of Action
- Proton pump inhibitor that suppresses gastric acid secretion
- Specific inhibition of the H+/K+-ATPase in the gastric parietal cell
- First single optical isomer of proton pump inhibitor, providing better acid control than racemic proton pump inhibitors
- Absorbed enterically
- Metabolized in the liver by the cytochrome P450 (CYP) enzyme system
- Plasma elimination half-life of approximately 1-1.5 hours

Recommended Dosage
- Healing of Erosive Esophagitis: 20 mg or 40 mg Once Daily for 4-8 Weeks
- Maintenance of Healing of Erosive Esophagitis: 20 mg Once Daily
- Symptomatic GERD: 20 mg Once Daily for 4 Weeks
- Helicobacter Pylori Eradication: Triple Therapy with Esomeprazole 40 mg Once Daily, Amoxicillin 1000 mg Twice Daily, Clarithromycin 500 mg Twice Daily for 10 days
- Zollinger-Ellison Syndrome: 20-80 mg Once Daily
- Acid-related Dyspepsia: 20-40 mg Once Daily for 2-4 weeks
- Duodenal Ulcer: 20 mg Once Daily for 2-4 weeks
- Gastric Ulcer: 20-40 mg Once Daily for 4-8 weeks
- Dosage adjustments for Pediatric & Elderly patients
Administration
- Tablet or capsule should be swallowed whole and taken one hour before a meal
- Oral Suspension requires mixing with water and administration within 30 minutes
- IV Injection should be administered over at least 3 minutes
Interaction
- Metabolized by CYP2C19 and CYP3A4
- No clinically significant interactions with certain drugs
- Potential interference with gastric pH-dependent drug absorption
Contraindications
- Known hypersensitivity to any of the formulation

- Use of antacids while taking Esomep

- No overall differences in safety and efficacy between elderly and younger individuals
- No dosage adjustment for patients with mild to moderate hepatic insufficiency
- Potential lack of alteration in pharmacokinetics in patients with renal impairment
