Indications
- Treatment of gastric and duodenal ulcer

Pharmacology
- Omeprazole, a substituted benzimidazole
- Inhibitor of gastric acid secretion
- Blocks hydrogen-potassium-adenosine triphosphatase
- Onset of antisecretory effect within one hour
- Maximum effect occurring within two hours
- Inhibition of secretion lasts up to 72 hours
- Secretory activity returns gradually over 3 to 5 days

Administration
- Direction for IV Injection administration

- Slow intravenous injection for IV Injection
- Intravenous infusion over a period of 20-30 minutes or more
- Reconstituted solution should not be mixed with any other drug
Interaction
- Decreased intragastric acidity affects ketoconazole absorption
- Metabolized in the liver through cytochrome P450
- Delayed elimination of diazepam, phenytoin, and warfarin
- Monitoring for patients receiving warfarin or phenytoin
- Increased plasma concentrations during concomitant administration with clarithromycin
- No evidence of an interaction with various drugs
Contraindications
- Known hypersensitivity to Omeprazole
- Gastric ulcer suspected with possibility of malignancy
Side Effects
- Skin rash, urticaria, pruritus
- Photosensitivity, bullous eruption, erythema multiforme
- Diarrhea, headache, constipation, nausea/vomiting
- Dizziness, light-headedness, somnolence, insomnia
- Reversible mental confusion, agitation, depression, hallucinations
- Arthritic and myalgic symptoms, blurred vision, taste disturbance
Pregnancy & Lactation
- No adverse effects on pregnancy or fetus/newborn child
- Can be used during pregnancy
- No information available on passage into breast milk
- Discontinue breastfeeding if Omeprazole use is considered essential
- Safety and effectiveness not established in pediatric patients less than 18 years of age
Precautions & Warnings
- Avoid concomitant use of clopidogrel and Omeprazole
- Increased risk for osteoporosis-related fractures
- Atrophic gastritis noted occasionally in long-term use
- Concomitant use with methotrexate may lead to toxicities
