Indications
- Relief from chronic heartburn symptoms
- Healing of erosive esophagitis

Pharmacology
- Proton pump inhibitor
- Suppresses gastric acid secretion
- Specific inhibition of the H+/K+-ATPase in the gastric parietal cell
- First single optical isomer of proton pump inhibitor
- Better acid control than racemic proton pump inhibitors
- Bioavailability is approximately 90%
- Extensively metabolized in the liver
- Plasma elimination half-life of approximately 1–1.5 hours
Dosage
- 20 mg or 40 mg Once Daily for 4-8 Weeks for erosive esophagitis
- 20 mg Once Daily for maintenance of healing of erosive esophagitis
- 20 mg Once Daily for symptomatic GERD
- 40 mg Once Daily in combination therapy for Helicobacter Pylori eradication
- 20-80 mg once daily for Zollinger-Ellison Syndrome
- 20-40 mg once daily for Acid-related Dyspepsia
- 20 mg once daily for Duodenal ulcer, 20-40 mg once daily for Gastric ulcer
- Dosage for IV Injection and IV infusion for different conditions and age groups
Administration
- Tablet or capsule should be swallowed whole and taken one hour before a meal
- Oral Suspension should be prepared and administered as per instructions
- IV Injection should be prepared and administered as per instructions
Interaction
- Metabolized in the liver by CYP2C19 and CYP3A4
- Inhibits gastric acid secretion
- Potential interference with absorption of specific drugs
- Clinically significant interactions with specific drugs
- Combination Therapy with Clarithromycin can affect plasma levels of Esonix and 14-hydroxyclarithromycin
Contraindications
- Known hypersensitivity to any of the formulation

Pregnancy & Lactation
- No adequate and well-controlled studies in pregnant women
- Animal studies reveal no teratogenic effects
- Excretion in milk not studied

- Symptomatic response to therapy does not preclude the presence of gastric malignancy
- Specific instructions for administration to patients with difficulty swallowing capsules

- Safety and effectiveness in paediatric patients not established
- No overall differences in safety and efficacy for geriatric use
- Dosage adjustment for severe hepatic insufficiency
- No alteration in Pharmacokinetics for renal impairment
Overdose Effects
- Lethal at high doses in animal studies
