Indications

Pharmacology
- Proton pump inhibitor
- Suppresses gastric acid secretion
- Inhibits the H+/K+-ATPase in the gastric parietal cell
- Better acid control than racemic proton pump inhibitors
- Enteric-coated pellet formulation
- Peak plasma levels occur at approximately 1.5 hours
- Extensively metabolized in the liver
- Plasma elimination half-life is approximately 1-1.5 hours

- Symptomatic response to therapy does not preclude the presence of gastric malignancy

- Use of antacids while taking esomeprazole
Use in Special Populations

- No overall differences in safety and efficacy observed in geriatric use
- No dosage adjustment recommended for patients with mild to moderate hepatic insufficiency
- No alteration in pharmacokinetics in patients with renal impairment
Overdose Effects
- Acute toxicity signs include reduced motor activity, changes in respiratory frequency, tremor, ataxia, and convulsions
- No specific antidote for esomeprazole
- Treatment should be symptomatic and supportive
