Description

  • Dopamine antagonist medication for the treatment of nausea and vomiting
  • Gastroprokinetic action based on blocking effect of dopamine receptors
  • Restores normal motility and tone of the upper gastrointestinal tract
  • Facilitates gastric emptying and enhances antral and duodenal peristalsis
  • Regulates contraction of the pylorus and increases esophageal peristalsis

Usability Features

  • Designed for oral administration before meals, if necessary, before retiring

Dosage

  • Adults: 10-20 mg (1-2 tablets or 10-20 ml suspension), every 6-8 hours daily
  • Children: 2-4 ml suspension/10 kg body weight or 0.4-0.8 ml pediatric drops/10 kg body weight, every 6-8 hours daily

Maximum Dosage

80 mg daily for adults

Indications

  • Dyspeptic symptom complex
  • Acute nausea and vomiting of various origins
  • Parkinson's disease induced nausea and vomiting
  • Speeding barium transit in follow-through radiological studies

Contraindications

  • Known hypersensitivity to domperidone
  • Neonates
  • Gastrointestinal hemorrhage, mechanical obstruction, or perforation
  • Prolactin releasing pituitary tumor (prolactinoma)

Side Effects

  • Transient intestinal cramps
  • Rare extrapyramidal phenomena in adults
  • Increase in plasma prolactin level
  • Rare allergic reactions such as rash and urticaria

Pregnancy & Lactation

  • Usage justified during first trimester if therapeutic benefit outweighs risk
  • Excreted in breast milk at lower levels compared to plasma concentrations
  • Nursing not recommended unless expected benefits outweigh potential risk

Precautions

  • Use with caution in children due to risk of extra-pyramidal reactions
  • Use with caution in patients with hepatic impairment

Special Populations

  • Use in infants with caution and under close medical supervision
  • Use with caution in patients with liver and kidney disorders
