Indications
- Relief from chronic heartburn symptoms
- Healing of erosive esophagitis
- Maintenance of healing of erosive esophagitis

Pharmacology
- Proton pump inhibitor
- Suppresses gastric acid secretion
- Enteric-coated pellet formulation
- Peak plasma levels occur at approximately 1.5 hours
- 97% bound to plasma proteins
- Extensively metabolized in the liver
- Plasma elimination half-life of approximately 1-1.5 hours
Dosage
- Dosage forms include oral tablets and IV injection
- Oral dosage for healing of erosive esophagitis: 20 mg or 40 mg once daily for 4-8 weeks
- IV injection or IV infusion for adult patients: 20 mg or 40 mg once daily
- Pediatric dosage based on body weight
Administration
- Tablet or capsule should be swallowed whole and taken one hour before a meal
- Oral suspension to be taken with water or administered through a nasogastric or gastric tube
- IV injection to be prepared with 0.9% Sodium Chloride
Interaction
- Metabolized by CYP2C19 and CYP3A4
- Inhibits gastric acid secretion
- May interfere with the absorption of certain drugs
Contraindications
- Contraindicated in patients with known hypersensitivity to any of the formulation
Side Effects
- Frequently occurring adverse events include headache, diarrhea, nausea, flatulence, abdominal pain, constipation, dry mouth
Pregnancy & Lactation
- No adequate and well-controlled studies in pregnant women
- Animal studies reveal no teratogenic effects
- Excretion in milk not studied
Precautions & Warnings
- Symptomatic response does not preclude the presence of gastric malignancy
- Guidelines for patients with difficulty swallowing capsules
Use in Special Populations
- No established safety and effectiveness in pediatric patients
- No significant differences in safety and efficacy between the elderly and younger individuals
- No dosage adjustment recommended for patients with mild to moderate hepatic insufficiency
Overdose Effects
- Lethal dosage observed in rats
- Major signs of acute toxicity include reduced motor activity, respiratory changes, tremor, convulsions

- Store at a temperature not exceeding 30°C in a dry place
- Protect from light and moisture
- Keep out of reach of children
