Composition
- 375 mg tablet: Amoxicillin 250 mg, Clavulanic Acid 125 mg
- 625 mg tablet: Amoxicillin 500 mg, Clavulanic Acid 125 mg
- 1 gm tablet: Amoxicillin 875 mg, Clavulanic Acid 125 mg
- Powder for Suspension: Amoxicillin 125 mg, Clavulanic Acid 31.25 mg
- Powder for Suspension (Forte): Amoxicillin 400 mg, Clavulanic Acid 57.5 mg
- 1.2 gm Injection: Amoxicillin Sodium 1 gm, Clavulanate Potassium 200 mg
- 0.6 gm injection: Amoxicillin Sodium 500 mg, Clavulanate Potassium 100 mg
Pharmacology
- Pharmacodynamic properties: Bactericidal activity against Gram-positive & Gram-negative microorganisms
- Pharmacokinetic properties: Optimized absorption at the start of a meal, low levels of serum binding
Dosage
- Adults and children over 12 years:
- 625 mg Tablet every 12 hours
- 375 mg Tablet every 8 hours
- 1 gm Tablet every 12 hours
- 1 gm Tablet every 8 hours
- Children 6-12 years:
- 2 teaspoonful every 8 hours
- 1 teaspoonful every 8 hours
- Children below 1 year: 25 mg/kg/day in divided doses every 8 hours
Administration
- Oral dosage form: May be taken without regard to meals, but absorption is enhanced when taken at the start of a meal
- IV injection: Administered intravenously by injection (over 2 minutes) or slow intravenous infusion (30 minutes)
Interaction
May reduce the efficacy of oral contraceptives, potential interaction with allopurinol
Contraindications
- History of Penicillin hypersensitivity
- Cross-sensitivity with other beta-lactam antibiotics
- Previous history of Co-amoxiclav or Penicillin-associated cholestatic jaundice
Side Effects
Uncommon and mainly of a mild and transitory nature, gastrointestinal side effects may be reduced by taking Co-amoxiclav at the start of meals
Pregnancy & Lactation
No teratogenic effect in animal studies, limited use in human pregnancy with no untoward effect, trace quantities detected in breast milk
Precautions & Warnings
Use with care in patients on anticoagulation therapy or with severe hepatic dysfunction
Use in Special Populations
Dose adjustment required in case of renal or hepatic impairment
Overdose Effects
Unlikely to occur, may cause gastrointestinal symptoms and electrolyte imbalances

Reconstitution
IV injection can be reconstituted by dissolving the powder in Water for Injection BP
Storage Conditions
Should be stored below 25°C, protected from light and moisture, reconstituted suspension should be kept in the refrigerator and used within 7 days
