Indications
- Short-term treatment of bacterial infections for upper respiratory tract infections, lower respiratory tract infections, genito-urinary tract infections, skin and soft tissue infections, bone and joint infections, and other infections

Pharmacology
- Pharmacodynamic properties: Amoxicillin and Clavulanic Acid combination. Broad spectrum bactericidal activity. Clavulanic acid inactivates a wide range of beta-lactamase enzymes.
- Pharmacokinetic properties: Peak serum levels occur about one hour after oral administration. Absorption is optimized at the start of a meal. Low levels of serum binding for both clavulanate and Amoxicillin.
Dosage
- Adults and children over 12 years:
- Tablet: 625 mg every 12 hours or 375 mg every 8 hours for usual dose.
- Suspension: Varies based on age and severity of infection.
- Forte suspension: Daily dosage varies based on infection severity.
- IV Injection: Dosage varies for adults and children based on age and severity of infection.
Administration
Oral dosage form can be taken without regard to meals. IV injection should not be administered intramuscularly or subcutaneously.
Interaction
May prolong bleeding time and prothrombin time. May reduce efficacy of oral contraceptives. Concomitant use with allopurinol may increase the likelihood of allergic skin reactions.
Contraindications
History of Penicillin hypersensitivity. Cross-sensitivity with other beta-lactam antibiotics. Also contraindicated for patients with previous history of Co-amoxiclav or Penicillin-associated cholestatic jaundice.
Side Effects
- Diarrhoea
- Pseudomembranous colitis
- Gastrointestinal side effects may be reduced by taking Co-amoxiclav at the start of meals.
- Hepatitis
- Cholestatic jaundice
- Urticarial and erythematous rashes
- Angioedema
- Anaphylaxis
Pregnancy & Lactation
Not recommended in pregnancy unless essential. Trace quantities can be detected in breast milk during lactation.
Precautions & Warnings
Use with care in patients on anticoagulation therapy or with severe hepatic dysfunction. Dosage should be adjusted for patients with moderate or severe renal impairment.
Use in Special Populations
Dose adjustment in case of patients with renal impairment or hepatic impairment.
Overdose Effects
Unlikely, may lead to gastrointestinal symptoms and fluid-electrolyte imbalances. May be removed from circulation by hemodialysis.

Reconstitution
IV injection can be reconstituted with Water for Injection BP. Should not be mixed with certain fluids for injection. Reconstituted solution may be injected into the drip tubing of infusion fluids over 3-4 minutes.
Storage Conditions
Should be stored below 25°C, protected from light and moisture. Reconstituted suspension should be refrigerated (not frozen) and used within 7 days. Reconstituted vial must be used within 20 minutes.
