Composition

  • 375 mg tablet: Amoxicillin 250 mg as Amoxicillin Trihydrate BP and Clavulanic Acid 125 mg as Diluted Potassium Clavulanate BP
  • 625 mg tablet: Amoxicillin 500 mg as Amoxicillin Trihydrate BP and Clavulanic Acid 125 mg as Diluted Potassium Clavulanate BP
  • 1 gm tablet: Amoxicillin 875 mg as Amoxicillin Trihydrate BP and Clavulanic Acid 125 mg Diluted Potassium Clavulanate BP
  • Powder for Suspension: Amoxicillin 125 mg as Amoxicillin Trihydrate BP and Clavulanic Acid 31.25 mg as Diluted Potassium Clavulanate BP
  • Powder for Suspension (Forte): Amoxicillin 400 mg as Amoxicillin Trihydrate BP and Clavulanic Acid 57.5 mg as Diluted Potassium Clavulanate BP
  • 1.2 gm Injection: Amoxicillin Sodium BP (equivalent to Amoxicillin 1 gm) and Clavulanate Potassium USP (equivalent to Clavulanic Acid 200 mg)
  • 0.6 gm injection: Amoxicillin Sodium BP (equivalent to Amoxicillin 500) and Clavulanate Potassium USP (equivalent to Clavulanic Acid 100 mg)

Pharmacology

  • Pharmacodynamic properties: Broad spectrum of bactericidal activity
  • Pharmacokinetic properties: Closely matched pharmacokinetics of the two components

Dosage

  • Adults and children over 12 years:
    • Tablet: 625 mg every 12 hours or 375 mg every 8 hours
    • Suspension: Varies with age and severity of infection
    • Forte suspension: Varies with age and severity of infection
    • IV Injection: Varies with age and severity of infection
  • Children of 2 to 12 years:
    • Mild to moderate infections: Suspension dosage varies with age
    • Serious infections: Forte Suspension dosage varies with age

Administration

Oral dosage form can be taken without regard to meals. IV injection for intravenous use only.

Interaction

  • Prolongation of bleeding time and prothrombin time
  • Reduction in efficacy of oral contraceptives
  • Concomitant use of allopurinol

Contraindications

History of Penicillin hypersensitivity or previous Co-amoxiclav or Penicillin-associated cholestatic jaundice

Pregnancy & Lactation

  • No teratogenic effect in animal studies
  • Limited use in human pregnancy with no untoward effect
  • Not recommended in pregnancy unless essential

Precautions & Warnings

  • Use with care in patients on anticoagulation therapy or with severe hepatic dysfunction
  • Dosage adjustment for moderate or severe renal impairment

Use in Special Populations

  • Dosage adjustment for renal impairment in adults and children
  • Dose with caution in hepatic impairment

Overdose Effects

  • Gastrointestinal symptoms
  • Disturbance of fluid and electrolyte balances
  • Potential removal by haemodialysis
