Indications
- Gastric and duodenal ulcer
- NSAID-associated duodenal and gastric ulcer
- Prophylaxis in patients with a history of NSAID-associated duodenal and gastric ulcer

Pharmacology
Inhibitor of gastric acid secretion by blocking hydrogen-potassium-adenosine triphosphatase (H+/K+ ATPase) enzyme system in the gastric parietal cell. Onset of the antisecretory effect occurs within one hour, with the maximum effect occurring within two hours and inhibition of secretion lasts up to 72 hours. Secretory activity returns gradually over 3 to 5 days upon discontinuation.
Dosage
- Oral dosage for various conditions
- IV injection for prophylaxis of acid aspiration, duodenal ulcer, gastric ulcer, reflux oesophagitis, Zollinger-Ellison syndrome
- Pediatric use in severe ulcerating reflux esophagitis
Administration
- IV injection should be given slowly over a period of 5 minutes
- Solution for IV injection obtained by adding water for injection to the vial containing powder
- IV infusion should be given over a period of 20-30 minutes
Interactions
- Decreased intragastric acidity may reduce the absorption of ketoconazole
- Metabolized in the liver through cytochrome P450, may delay elimination of diazepam, phenytoin, and warfarin
- Concomitant treatment with Seclo can affect plasma concentrations of other drugs
Contraindications
Contraindicated in patients with known hypersensitivity to omeprazole
Side Effects
- Mild and reversible adverse reactions
- Skin rash, urticaria, pruritus, photosensitivity, bullous eruption, erythema multiforme, angioedema, alopecia
- Diarrhoea, headache, constipation, nausea/vomiting, flatulence, abdominal pain
- Dry mouth, stomatitis, candidiasis, paraesthesia, dizziness, light headedness, somnolence, insomnia, vertigo
- Reversible mental confusion, agitation, depression, hallucinations, arthritic and myalgic symptoms
- Blurred vision, taste disturbance, peripheral edema, increased sweating, gynaecomastia, hematological abnormalities, hepatic failure, increases in liver enzymes
Pregnancy & Lactation
No adverse effects on pregnancy or on the health of the fetus/newborn child with use of omeprazole. Use during pregnancy is acceptable. Discontinue breastfeeding if essential.
Precautions & Warnings
- Avoid concomitant use of clopidogrel and Seclo
- PPI therapy may be associated with increased risk for osteoporosis-related fractures
- Atrophic gastritis noted occasionally in patients treated long-term with Seclo
- Concomitant use of PPIs with methotrexate may lead to methotrexate toxicities
