Pharmacology

  • Mode of Action: Proton pump inhibitor that suppresses gastric acid secretion
  • Absorption: Peak plasma levels occur at approximately 1.5 hours after oral administration
  • Distribution: 97% bound to plasma proteins
  • Metabolism: Extensively metabolized in the liver by the cytochrome P450 enzyme system
  • Excretion: Plasma elimination half-life is approximately 1–1.5 hours

Dosage

  • Oral Dosage Form:
    • Healing of Erosive Esophagitis: 20 mg or 40 mg Once Daily for 4-8 Weeks
    • Symptomatic GERD: 20 mg Once Daily for 4 Weeks
    • Helicobacter Pylori Eradication: Triple Therapy with Esomeprazole 40 mg Once Daily for 10 days
  • IV Injection or IV Infusion: Dosage varies for adult and pediatric patients

Administration

  • Tablet or Capsule: Should be swallowed whole and taken one hour before a meal
  • Oral Suspension: Should be taken with water and consumed within 30 minutes
  • IV Injection: Must be administrated intravenously over a period of at least 3 minutes

Interaction

  • Metabolism: Extensively metabolized in the liver by CYP2C19 and CYP3A4
  • Inhibition: Potential interference with the absorption of drugs where gastric pH is essential for bioavailability

Contraindications

Contraindicated in patients with known hypersensitivity to any of the formulation

  • No overall differences in safety and efficacy observed between the elderly and younger individuals

Overdose Effects

Lethal dose in rats, may cause reduced motor activity, respiratory changes, tremor, and convulsions

Storage Conditions

Store at a temperature not exceeding 30°C in a dry place, protect from light and moisture
