Indications

  • Relief from chronic heartburn symptoms
  • Healing of erosive esophagitis
  • Maintenance of healing of erosive esophagitis
  • Eradication of Helicobacter pylori infection
  • Zollinger-Ellison Syndrome
  • Treatment of acid related Dyspepsia
  • Management of Duodenal & Gastric ulcer

Pharmacology

  • Proton pump inhibitor
  • Suppresses gastric acid secretion
  • Enteric-coated pellet formulation
  • Peak plasma levels occur at approximately 1.5 hours after oral administration

Dosage

  • Healing of erosive esophagitis: 20 mg or 40 mg Once Daily for 4-8 Weeks
  • Maintenance of Healing of Erosive Esophagitis: 20 mg Once Daily
  • Symptomatic GERD: 20 mg Once Daily for 4 Weeks
  • Helicobacter Pylori eradication: Triple Therapy for 10 days
  • Zollinger-Ellison Syndrome: 20-80 mg Once Daily
  • Acid-related Dyspepsia: 20-40 mg Once Daily for 2-4 weeks
  • Duodenal ulcer: 20 mg Once Daily for 2-4 weeks
  • Gastric ulcer: 20-40 mg Once Daily for 4-8 weeks
  • IV Injection for Adults: 20 mg or 40 mg once daily
  • IV Injection for Pediatric Patients
  • Risk reduction of rebleeding of Gastric or Duodenal Ulcers: 80 mg

Administration

  • Tablet or capsule should be swallowed whole and taken one hour before a meal
  • Oral Suspension should be taken after mixing with water
  • IV Injection should be prepared by adding 0.9% Sodium Chloride for intravenous injection

Interaction

  • Metabolized by CYP2C19 and CYP3A4
  • No clinically relevant interactions with specific drugs
  • Potential interference with CYP2C19
  • May interfere with the absorption of certain drugs

Contraindications

  • Contraindicated in patients with known hypersensitivity to any of the formulation

Pregnancy & Lactation

  • No adequate and well-controlled studies in pregnant women
  • Animal studies have revealed no teratogenic effects

  • Breastfeeding should be discontinued if essential

Precautions & Warnings

  • Symptomatic response to therapy does not preclude the presence of gastric malignancy
  • Instructions for patients regarding capsule intake
  • Antacids may be used while taking the medication

Use in Special Populations

  • Safety and effectiveness in pediatric patients not established
  • No overall differences in safety and efficacy observed between the elderly and younger individuals
  • No dosage adjustment recommended for patients with mild to moderate hepatic insufficiency
  • Pharmacokinetics not expected to be altered in patients with renal impairment

Overdose Effects

  • Acute toxicity in rats
  • No specific antidote known
  • Treatment should be symptomatic and supportive
