Indications

Gastric and duodenal ulcer

Pharmacology

Inhibitor of gastric acid secretion, blocks hydrogen-potassium-adenosine triphosphatase (H+/K+ ATPase) enzyme system, onset of effect within one hour, maximum effect within two hours, inhibition lasts up to 72 hours, gradual return of secretory activity over 3 to 5 days after drug discontinuation

Interaction

Decreased intragastric acidity affecting ketoconazole absorption
Metabolism through cytochrome P450 leading to delayed elimination of diazepam, phenytoin, and warfarin
Plasma concentrations affected during concomitant administration with clarithromycin

Contraindications

Known hypersensitivity to omeprazole, suspicion of gastric ulcer malignancy

Side Effects

Mild and reversible adverse reactions including skin rash, urticaria, pruritus, photosensitivity, bullous eruption, diarrhea, headache, constipation, nausea/vomiting, dizziness, somnolence, insomnia, arthritic and myalgic symptoms, visual disturbances, taste disturbance, peripheral edema, gynaecomastia, hematological reactions, anaphylactic shock, hepatic reactions

Pregnancy & Lactation

Use during pregnancy deemed safe, no information on potential effects in breast milk or on neonate

Precautions & Warnings

Avoid concomitant use with clopidogrel
Possible association with increased risk for osteoporosis-related fractures
Atrophic gastritis observed in long-term use
Potential interaction with methotrexate
