Indications
- Gastric and duodenal ulcer
- NSAID-associated duodenal and gastric ulcer
- Prophylaxis in patients with a history of NSAID-associated ulcers
- Gastro-esophageal reflux disease
- Long-term management of acid reflux disease
- Acid-related dyspepsia
- Severe ulcerating reflux esophagitis
- Prophylaxis of acid aspiration during general anesthesia
- Zollinger-Ellison syndrome

Pharmacology
- Inhibitor of gastric acid secretion
- Blocks hydrogen-potassium-adenosine triphosphatase (H+/K+ ATPase) enzyme system
- Onset of antisecretory effect within one hour
- Maximum effect occurring within two hours
- Inhibition of secretion lasts up to 72 hours
Dosage
- 20 mg once daily for benign gastric and duodenal ulcer
- 40 mg daily for severe or recurrent cases
- 8 weeks in gastric ulceration
- Maintenance dose for recurrent duodenal ulcer: 20 mg once daily
- 10-20 mg daily for prevention of relapse in duodenal ulcer
- 10-20 mg once daily for acid-related dyspepsia
- 40 mg on the preceding evening for prophylaxis of acid aspiration
- 20-120 mg daily for Zollinger-Ellison syndrome
Administration
- Omeprazole IV injection for intravenous administration only
- Omeprazole IV infusion as an intravenous infusion
- Omeprazole IV infusion given over a period of 20-30 minutes or more
Interaction
- Decreased intragastric acidity affecting absorption of ketoconazole
- Metabolism through cytochrome P450 affecting elimination of certain drugs
- Plasma concentrations of Omeprazole and clarithromycin increased during concomitant administration
Contraindications
- Known hypersensitivity to Omeprazole
- Suspected gastric ulcer with possibility of malignancy
Side Effects
- Skin rash, urticaria, pruritus
- Photosensitivity, bullous eruption, erythema multiforme
- Diarrhoea, headache
- Constipation, nausea/vomiting, flatulence, abdominal pain
- Paraesthesia, dizziness, light headedness, feeling faint
- Dry mouth, stomatitis, candidiasis
Pregnancy & Lactation
- No adverse effects on pregnancy or fetus/newborn child observed
- Consideration for use during pregnancy
- No information available on passage into breast milk or neonate
Precautions & Warnings
- Avoid concomitant use of clopidogrel and Zilon
- Association of PPI therapy with increased risk for osteoporosis-related fractures
