Indications

Pharmacology
- Proton pump inhibitor
- Suppresses gastric acid secretion
- Inhibits H+/K+-ATPase
- Provides better acid control than racemic proton pump inhibitors
- Enteric-coated pellet formulation of esomeprazole magnesium
- Peak plasma levels occur at approximately 1.5 hours after oral administration
- Metabolized in the liver by the cytochrome P450 (CYP) enzyme system
- Plasma elimination half-life of approximately 1–1.5 hours
Dosage
- Oral dosage form
- Dosage varies for different indications
- 20 mg or 40 mg once daily for healing of erosive esophagitis
- 20 mg once daily for maintenance of healing of erosive esophagitis
- 20 mg once daily for symptomatic GERD
- Triple Therapy for Helicobacter pylori eradication
- Dose varies for Zollinger-Ellison Syndrome, Acid-related Dyspepsia, Duodenal & Gastric ulcer
- IV injection or IV infusion for certain indications
Administration
- Tablet or capsule should be swallowed whole and taken one hour before a meal
- Specific instructions for oral suspension administration
- Preparation of solution for IV injection
- Recommendation for administration through a nasogastric or gastric tube
Interaction
- Extensively metabolized in the liver by CYP2C19 and CYP3A4
- No clinically relevant interactions with certain CYP enzymes
