Indications

  • Relief from chronic heartburn symptoms
  • Healing of erosive esophagitis
  • Maintenance of healing of erosive esophagitis
  • Eradication of Helicobacter pylori infection
  • Zollinger-Ellison Syndrome treatment
  • Treatment of acid-related Dyspepsia
  • Treatment of Duodenal & Gastric ulcer

Pharmacology

  • Proton pump inhibitor
  • Suppresses gastric acid secretion
  • Inhibition of the H+/K+-ATPase
  • Better acid control than racemic proton pump inhibitors
  • Enteric-coated pellet formulation
  • Extensively metabolized in the liver
  • Cytochrome P450 (CYP) enzyme system dependent metabolism
  • Plasma elimination half-life of approximately 1-1.5 hours

Dosage

  • Oral dosage form
  • IV Injection or IV infusion
  • Variety of doses based on indication and patient age/weight
  • Specific dosages for different indications such as zollinger-ellison syndrome and Helicobacter pylori eradication

Administration

  • Tablet or capsule should be swallowed whole and taken one hour before a meal
  • Oral suspension should be taken with specific instructions on mixing and administration

Interaction

  • Metabolized by CYP2C19 and CYP3A4

Overdose Effects

  • Lethal at high doses in animal studies
  • Reduced motor activity, changes in respiratory frequency, tremor, and convulsions
