Indications
- Gastric and duodenal ulcer
- NSAID-associated duodenal and gastric ulcer
- Gastro-esophageal reflux disease
- Acid-related dyspepsia
- Severe ulcerating reflux esophagitis
- Zollinger-Ellison syndrome
- Helicobacter pylori-induced peptic ulcer
Pharmacology
Inhibitor of gastric acid secretion
Dosage
- 20 mg once daily for benign gastric and duodenal ulcer
- 20 mg once daily for NSAID-associated duodenal or gastric ulcer
- 20 mg once daily for gastro-esophageal reflux disease
- 10-20 mg daily for long-term management of acid reflux disease
- 10-20 mg once daily for acid-related dyspepsia
- 40 mg on the preceding evening for prophylaxis of acid aspiration
- Initially 60 mg once daily for Zollinger-Ellison syndrome
- Pediatric use: 10-40 mg once daily depending on body-weight

Interaction
- Decreased absorption of ketoconazole
- Metabolism in the liver may delay elimination of diazepam, phenytoin, and warfarin
- Plasma concentrations increased during concomitant administration with clarithromycin
- Increased bioavailability of digoxin during simultaneous treatment

Known hypersensitivity
Side Effects
- Skin rash, urticaria, pruritus
- Diarrhoea, headache
- Gastrointestinal reactions: constipation, nausea/vomiting, flatulence, abdominal pain
- Dizziness, somnolence, insomnia, confusion, depression
- Arthritic and myalgic symptoms
- Increases in liver enzymes
Pregnancy & Lactation
- Pregnancy: No adverse effects indicated
- Lactation: Discontinuation of breast-feeding recommended
Precautions & Warnings
- Avoid concomitant use with clopidogrel
- Potential association with increased risk for osteoporosis-related fractures
- Atrophic gastritis observed in long-term treatment
